The treatment with all-trans retinoic acid causes apoptosis without chromosomal instability in adipose-derived stem cells and might act starting the browning process

Introduction: The knowledge about the biology of obesity and apoptosis induction adipose-derived stem cells (ADSCs) may support in development new therapies. Objective: present research aimed to investigate toxicogenic effect all-trans retinoic acid (ATRA) as well its influence adipogenic differentiation expression genes related DNA damage, cell cycle thermogenesis. Methodology: Cell cultures human were treated for 12 hours with ATRA (20.75 μM) performed differentiation, comet assay, micronucleous, death, qPCR. Results: treatment decreased capacity ADSCs. assay demonstrated increase nucleoid frequency genomic damage scoring. However, these damages not fixed at chromosome level, since tail moment was significant. increased cytotoxicity death by apoptosis. relative CHEK-1, CHEK-2, CDC25A, CDC25C, ATM ATR only UCP1 significantly. Conclusion: results study demonstrate that use induces ADSCs, although it eliminated process. Therefore, administration ADSCs did cause chromosomal instability, which suggests safety. Also considered might active browning effect. Thus, this compound is a promising candidate novel therapies / or localized fat mesotherapy.
